According to most Rishonim, Sefirat ha'Omer is a זכר למקדש (per אמימר in מנחות סו) and, as such, is דרבנן in our non-Mikdash era. Rambam differs and reckons the Mitzvah as being "in force" today as it was in the times that an Omer was offered on the Mizbeach.
